When you arrive at Sandgem Town, Prof. Rowan's assistant will meet you at the entrance and escort you to the research lab. You'll run into your friend again.
When you confront Prof. Rowan, he will see that you treated your Pokemon well, so he will let you keep it, and even rename it if you wish. He will ask you for a favor in return: to travel through Sinnoh and collect info of all of the Pokemon in a Pokedex. You now have the Pokedex, and the ability to record all the info of the Pokemon you will encounter.
When you leave the lab, Prof. Rowan's assistant will take you on a little tour of the town and what all it has to offer, which isn't much. You will first pass by the Pokemon Center, where you can heal your Pokemon for free, so be sure to check out that place a lot. After that, you will pass by the Pokemart, where you can buy various items to help you along the way. Every Pokemart in each town is different in terms of what items they sell, so be sure to check out all of them. After you pass by the Pokemart, the assistant will send you home to tell your mom about your new job.
After you visit your mom, you will find Prof. Rowan's assistant waiting for you at the edge of the grass on Route 202. She (or he if you picked a female trainer) will show you how to catch wild Pokemon and give you five Pokeballs to get you started.
Be sure to heal your Pokemon at the Pokemon Center before you go on if need be, because you will encounter trainers where you will need to battle. After you heal, head north to arrive at Jubilife City.

Diamond and Pearl part 1

The game starts with Prof. Rowan explaining the game to you. You will have the option to choose between a male or female character, and you will be able to name your character and your rival. Your adventure offically starts in Twinleaf Town, your character's home town.
As much as you may like to hang out in your bedroom playing with your Wii, you need to get started. Check the computer and a note on the wall for some game info, then go downstairs to talk to your character's mom. She will explain that your friend (rival) has called and wants you to visit him at his house (northwest building).
Apparently you took too long, so your friend leaves and forgets his bag and journal. When he comes back, follow him upstairs. He'll grab his stuff and ask you to follow him. Go north toward Route 201 to find him.
When you meet up with him, he'll explain that he's heading to Lake Verity to search for unique Pokemon. Head west down to Route 201 and enter Lake Verity via the north path through the trees.
When you enter, you will pass by Prof. Rowan and his assistant. When you investigate the briefcase that they left behind, you will be confronted by Starly, so you will have no choice but to grab a Pokemon from one of the three Pokeballs. The Pokemon you choose will be your primary Pokemon for the rest of the game. After the battle, return to Twinleaf.
Pokemon to choose from:
Turtwig

A grass type that will gain a ground type when it evolves. Good for beginners.
Chimchar

A fire type that will gain a fighting type when it evolves. Good for experts.
Piplup

A water type that will gain a steel type when it evolves. Good for intermediate players.
On your way back home, you will run into Prof. Rowan and his assistant. After he finds out you used his Pokemon, he'll take off. His assistant will tell you to visit the Prof. in Sandgem Town, but go back to Twinleaf Town first. When you arrive back home, talk to your mom to explain your little incident. She'll tell you to go visit Prof. Rowan in Sandgem Town. She'll then give you a pair of running shoes, to speed things up a bit (to use the shoes, simply hold down B while you walk). To get to Sandgem Town, head east through Route 201.

New walkthrough

The new walk-through is coming up is of Diamond and Pearl and not lke the others this is going to be a written one. I am warning you to stay tuned.
